اثر عصارههای سیر و هل بر روی خواص مورفولوژی و فیزیولوژی استافیلوکوکوس اورئوس مقاوم به متی سیلین (MRSA) و سودوموناس آئروجینوزا

<div style="text-align: justify;"><strong>مقدمه:</strong> سیر با نام علمی <em>Allium sativum</em> متعلق به خانواده Liliaceae و هِل با نام علمی <em>Elettaria Cardamom</em> Maton متعلق به خانواده Zingiberaceae دارای کاربردهای درمانی در طب سنتی هستند که برخی از آنها با بررسیهای علمی اثبات شده است. از این دو گیاه اثرات ضدمیکروبی گزارش شده است. <strong>هدف:</strong> هدف از این تحقیق، بررسی اثر عصاره آبی سیر و عصاره متانولی هل بر روی سوشهای مقاوم استافیلوکوکوس اورئوس و سودوموناس آئروجینوزا است. <strong>روش بررسی:</strong> ابتدا حبههای سیر (100گرم) پوست گرفته شده و در آب مقطر استریل به نسبت 1 به 1 در مخلوط کن هموژن شدند، پس از سانتریفیوژ محلول رویی سیر جمعآوری گردید و از میان یک کاغذ صافی و در نهایت از فیلتر میلی پور (45/0 میکرون) عبور داده شد و برای انجام آزمایشها در یخچال (4 درجه سانتیگراد) نگهداری شد. برای تهیه عصاره هل، پس از جمعآوری دانههای هل، خشک کردن و پودر کردن آنها، عصارهگیری با استفاده از متانول و با روش پرکولاسیون انجام گرفت. برای بررسی اثرات ضدمیکروبی عصارههای سیر و هل، سوسپانسیون میکروبی با کدورتی مطابق با 5/0 مک فارلند تهیه و رقتهای مختلف عصارهها شامل (v/v 1:64... 1:8، 1:4، 1:2 برای سیر و رقتهای µg/ml 50، ... ، 400، 650، 800 برای هل) تهیه و علیه سویههای 33591 ATCC MRSA و ATCC 27853 P. aeroginosa آزمایش شد. حداقل غلظت متوقفکننده رشد از طریق روشهای دیسکگذاری، چاهک و رقت در لوله تعیین شد. همچنین در این بررسی تغییرات مورفولوژی MRSA و <em>P. aeroginosa</em> با استفاده از میکروسکوپ نوری و خصوصیات فیزیولوژی آنها با استفاده از تلقیح غلظتهای کمتر از بازدارنده سیر و هل به کشت باکتریایی انجام شد. <strong>نتایج:</strong> عصاره هل، هیچ اثری بر روی سو دوموناس آئروجینوزا نداشت، اما عصاره سیر بر روی این باکتری موثر بود. عصاره سیر (رقتهای 1:2،1:4،1:8،1:16 حاوی µg/ml 5/27، 110، 55، 220 آلیسین) رشد MRSA را مهار میکرد و غلظتهای (v/v) 1:8 تا 1:2 آن باعث مهار رشد سودوموناس آئروجینوزا میشد. همچنین دیده شد که MRSA به عصاره هل در رقتهای µg/ml 200 تا 800 حساس است. به طور کلی حداقل غلظت مهارکننده سیر برای MRSA (MIC سیر =1:16: مقدار آلیسین g/mlµ 50/27) کمتر از حداقل غلظت مهارکننده سیر برای سودوموناس آئروجینوزا (MIC سیر =1:8 و مقدار آلیسین µg/ml 55) است بررسی اثر عصاره سیر بر روی مورفولوژی باکتریها شامل تغییر اندازه آنها، تخریب شکل جزئی و تورم سلولی است اما عصاره هل هیچ تغییر شکل آشکاری را بر روی سلول در مقایسه با کنترل نشان نمیدهد. همچنین در غلظتهای پایین سیر، تولید آنزیم کاتالاز و پیگمان پیوسیانین توسط سودوموناس آئروجینوزا کاهش یافت اما توانایی همولیز آن افزایش پیداکرد. نتایج نشان داد که تولید اسید از ساکارز و مانیتول توسط MRSA بر خلاف شاهد، منفی شد. <strong>نتیجهگیری:</strong> این بررسی نشان داد که عصاره سیر، اثرات ضدباکتریایی متفاوتی را علیه 2 باکتری مورد آزمایش اعمال میکند.</div>
<div style="text-align: justify;"><strong>Background: </strong>Garlic has been known as an important medicinal plant for centuries and belonging to the Liliaceae family. Cardamom is the dried fruit of the tall perennial herbaceous plant, <em>Elettaria cardamomum</em> Maton, and belonging to the Zingiberaceae. family. <strong>Objective:</strong> The purpose of this study was to investigate the effect of aqueous extract of garlic and methanolic extract of cardamom on MRSA and <em>P. aeruginosa</em>. <strong>Method: </strong>Dry garlic bulbs (100g) were peeled and homogenized distilled water (1:1 w/v), using a blender and 50% juice of garlic is obtained, centrifuged and then the supernatant of garlic juice is collected and passed through a 0.45µm filter and stored at 4˚c for further experiments. Also after collecting cardamom seeds, drying and making it powder, extracts were obtained by using percolation method with methanol. for evaluating the antibacterial activity of the extracts of garlic and cardamom, the microbial suspension was prepared by direct colony suspension method and different dilutions of extracts (1:2, 1:4, 1:8, …, 1:64 v/v for garlic 800, 650, 400, …, 50 µg/ml for cardamom) were prepared and tested against of MRSA ATCC 33591 and <em>P. aeruginosa</em> ATCC 27853. Minimum Inhibition Concentration (MIC) was obtained via Disc diffusion and Broth dilution and well diffusion method. We also analyzed morphological changes of MRSA and <em>P.aeruginosa</em> by light microscopy (LM) and biochemical properties was studied by inoculation of low concentrations of garlic and cardamom extracts to bacterial culture. <strong>Results:</strong> The results showed that cardamom extract has no effect against Pseudomonas aeruginosa but garlic extract is effective on <em>P. aeruginosa</em>. Garlic extract (1:2, 1:4, 1:8, 1:16, containing 220, 110, 55, 27.50 µg/ml allicin) inhibited the growth of MRSA and concentrations of 1:2 to 1:8 (v/v) inhibited the growth of P<em>seudomonas aeruginosa</em>. Further more MRSA is sensitive to cardamom extract in 800 to 200µg/ml. In general, the minimal inhibitory concentration for MRSA (garlic MIC 1:16 allicin mean MIC 27.50 µg/ml) were lower than for <em>P. aeruginosa</em> (garlic MIC 1:8 allicin mean MIC 55µg/ml).The treatment group with garlic extract showed a changed form of morphology such as cellular swelling, partially distored shape and changes in the size of bacteria, but cardamom extract dose not reveal any changes compared to the control. It was also observed in low concentrations of garlic, production of catalase enzyme and pyocyanine pigment by <em>P.aeruginosa</em> were decreased but it increased strain hemolysis ability, and acid production from saccharose, manitol by MRSA changed to negative. <strong>Conclusion:</strong> This research showed that garlic and cardamom extracts have different antibacterial properties against 2 tested bacteria.</div>
