بررسی تاثیر عصاره <i>Scutellaria pinnatifida</i> در مهار سمیت سلولی تجمعات سمی آلفاسینوکلئین

<div style="text-align: justify;">بسیاری از بیماری­های تخریب سیستم عصبی با تشکیل تجمعات پروتئینی موسوم به فیبریل های آمیلوئیدی همراه­اند. تجمعات آمیلوئیدی آلفا سینوکلئین به عنوان عامل کلیدی در بروز و گسترش بیماریهای تحلیل نورونی در مغز شناخته شده است و یافتن ترکیباتی که بتوانند تجمعات و یا سمیت آنها را مهار کنند در کانون توجه مطالعات در زمینه بیماری های تخریب سیستم عصبی هستند. در این مطالعه اثرات عصاره های استخراجی از ریشه گونه <em><span dir="LTR">S. pinnatifida) Scutellaria pinnatifida</span></em>) بر روی مهار سمیت نورونی گونه های تجمعی- سمی آلفا سینوکلئین بررسی شد. به این منظور فرایند فیبریلاسیون در آلفاسینوکلئین القا و با روش¬های استاندارد مانند سنجش فلورسنس ترکیب تیوفلاوین تی و مطالعه تصاویرمیکروسکوب فلورسنس تایید شد. میزان سمیت نورونی تجمعات سمی فیبریلی آلفاسینوکلئین با تیمار رده سلولی <span dir="LTR">PC12</span> و با روشهای <span dir="LTR">MTT</span> و <span dir="LTR">LDH </span> سنجیده شد. نتایج نشان دهنده­ی مرگ معنی دار در سلولهای تیمار شده با آلفاسینوکلئین بود. برخلاف استخراج های متانولی، هگزانی و اتیل استات <em><span dir="LTR">S. pinnatifida</span></em> استخراج های دی کلرومتان و بوتانل نرمال مانع مرگ سلولی در حضور آلفا سینوکلئین شدند. همچنین مشخص گردید که استخراج های دی کلرومتان و بوتانل نرمال حاوی میزان قابل توجهی فلاونوئید با خاصیت آنتی­اکسیدانی بالا می­باشند. با توجه به مطالعه صورت گرفته میتوان نتیجه گرفت که عصاره های استخراجی خاصی از <em><span dir="LTR">S. pinnatifida</span></em> تاثیرات مخرب تجمعات سمی پروتئین آلفا سینوکلیئن را مهار می­کنند و می­توانند برای مهار انتقال و گسترش آسیب های وابسته به تجمعات آلفا سینوکلئین مورد توجه باشد که این امر نیازمند تحقیقات بیشتر بر روی ترکیبات موجود در این گیاه می­باشد.</div>
<div style="text-align: justify;">Many of neurodegenerative diseases (NDs) are associated with formation of the protein aggregates called amyloidal fibrils. Fibrillization of alpha-synuclein seems to be a key stage in the creation and progression of NDs and finding the compounds that inherent the toxicity of alpha-synuclein aggregates seems to be one of the most attention subjects in the neurodegenerative studies. In this study, the effects of various extracts derived from the roots of<em> S. pinnatifida, on</em> the alpha-synuclein cytotoxicity were assayed. In this regard, the process of fibrillization was induced in alpha-synuclein and then formation of aggregates was verified using standard methods such as thioflavin T fluorescence assay and microscopic fluorescence images. Neurotoxicity of alpha-syncline fibrils on PC12 cells was examined by MTT and LDH methods. Results showed that, after treatment with alpha-synuclein aggregates, the rate of cell death was evaluated significantly. In spite of the methanolic, hexane and ethyl acetate extracts, in the presence of dichloromethane and n.butanol extracts, the death of cells treated with alpha-synuclein were inhibited significantly. Furthermore, it was found that dichloromethane and n.butanol extracts contained high amount of anti-oxidant flavonoids. It can be concluded that the extracts of <em>S. pinnatifida</em>, significantly inhibit the harmful effects of toxic alpha-synuclein aggregates and can be used to treat amyloid-related disorders. However, it is required more investigations on the kind of the effective compounds of <em>S.</em><em> pinnatifida</em> extracts.<span dir="RTL"></span></div>
