مقدمه: لیشمانیوز جلدی (سالک) توسط گونه‌های مختلف لیشمانیا ایجاد می‌شود. ترکیبات پنج ظرفیتی آنتیموان به عنوان خط اول درمان سالک، همراه با محدودیت‌ها و عوارض جانبی می‌باشند. استفاده از داروهای گیاهی به دلیل سمیت و هزینه کمتر-کارایی و دسترسی بیشتر مورد توجه قرار دارند. هدف: هدف از این مطالعه بررسی اثر عصاره هیدروالکلی ریشه شیرین‌بیان و گلیسیریزیک ‌اسید بر پروماستیگوت‌ها و آماستیگوت‌های لیشمانیا ماژور می‌باشد. روش بررسی: عصاره هیدروالکلی ریشه شیرین‌بیان، گلیسیریزیک ‌اسید یا گلوکانتیم در دوزهای مختلف همراه با پروماستیگوت لیشمانیا ماژور در دمای ٢٦ درجه سانتی‌گراد در پلیت کشت سلول به مدت ٢٤ و ٤٨ ساعت انکوبه و درصد بقا پروماستیگوت‌ها به روش MTT بررسی شد. سپس دوزهای مشخص از عصاره هیدروالکلی ریشه شیرین‌بیان، گلیسیریزیک ‌اسید یا گلوکانتیم به کشت ماکروفاژهای آلوده به انگل لیشمانیا ماژور اضافه و بعد از ٢٤ ساعت میانگین تعداد آماستیگوت‌ها در ماکروفاژها محاسبه شد. نتایج: نتایج حاصل از درصد بقا پروماستیگوت و آماستیگوت لیشمانیاماژور نشان داد که IC٥٠ عصاره هیدروالکلی ریشه شیرین‌بیان، گلیسیریزیک ‌اسید و گلوکانتیم بر پروماستیگوت به‌ ترتیب ٠١٨/٠ ± ١٢٥٠، ٠١٧/٠±٣٠٠٠ و ٠٤٣/٠±٥٠ میکروگرم در میلی‌لیتر بعد از ٢٤ ساعت و ٠١٦/٠ ± ٠١٧/٠‌، ١٠٠٠/٠ ± ٣٠٠٠ و ٠٠٩/٠ ± ٢٥ میکروگرم در میلی‌لیتر بعد از ٤٨ ساعت می‌باشد. همچنین IC٥٠ عصاره ریشه، گلیسیریزیک ‌اسید و گلوکانتیم بر آماستیگوت لیشمانیا ماژور به ‌ترتیب ٥٠٠، ١٠٠٠ و ٢٥ میکروگرم در میلی‌لیتر برآورد شد. نتیجه‌گیری: عصاره هیدروالکلی ریشه شیرین‌بیان و گلیسیریزیک ‌اسید دارای اثر کشندگی بر پروماستیگوت و آماستیگوت ‌لیشمانیا ماژور در شرایط برون‌تنی است.
